Yes, you can absolutely take a manual backup at your convenient time apart from the nightly automated backup that WPTC does.

You can do this by going to your WordPress dashboard -> WP Time Capsule -> Settings -> Start Backup.

Note: If you want to stop an existing backup, you will also see a Stop Backup button available in place of the Start Backup button.
